Sistemas injectáveis para a regeneração óssea Another example of injectable systems under development TE strategy to deliver bone cells The whole idea is to immobilize cells whithin a biodegrad polym microsphere While polym degrades, cells multiply and synthesize ECM Eventually the polym disapears from site, leaving new tissue Polímero Célula Matriz extracelular
Rowley et al, Biomaterials (1999) Alginato Rowley et al, Biomaterials (1999) The alginate modification consisted in three different processes: the irradiation, the oxidation and the RGD coupling to the polymer chains. The irradiation will decrease the molecular weight of alginate molecule, which willlead to a decrease in the viscosity of the solution. This will be an advantage in the cell/polymer mixing step. If the polymer has a high viscosity the cells will be exposed to high shear forces which could lead to a decrease in cell viability. After, the alginate was oxidized to increase the biodegradability. Oxidation of alginate to cleave the carbon-carbon bond of the cis-diol within ssacharide rings also decreased the total Mw of alginates in proportion to the molar ratio between NaIO4 added to the reaction and the number of alginate residues in the solution. Oxidized low Mw MVG k(degradadtion constant rate) = 18x10-3/h high Mw MVG k=5x10-3/h NOTE (alsberg, 2003) 5Mrads - it provides ultimate clearence by kidneys. Irradiates alginate at or below 50kDa. http://www.steve.gb.com/science/extracellular_matrix.html
