Assuntos Regulatórios - Fitoterápicos
FITOTERÁPICO = MEDICAMENTO Legislação aplicável - no geral, toda a legislação referente a medicamentos, + regulamento específico para registro -> RDC 48/04, e RE’s 88, 89, 90 e 91/04.
Polar Solvent Extract from the Euphorbiaceae Plants
The standardized extract of the butanolic fraction of the Euphorbia tirucalli latex, has antitumoral and citotoxic action; The formulation of this extract contains 0,001% to 95% of the active fraction extracted from the plant latex and an adequate ammount is between 0.1 mg to 2000 mg, particularly, between 10 mg and 100 mg of the compounds. Dagoberto C. Brandão PHC Pharma Consulting

Polar Solvent Extract from the Euphorbiaceae Plants
During tests to verify its anticarcinogenic activity in cultures of carcinogenic cells from the lungs, breast, colon, kidneys, ovary and prostate, it was revealed as active, since there was inibition of 50% of growth. CI50 values (inibitory concentration able to produce 50% of the maximum effect) show the efficacy of the active fraction from the latex extract as anti-carcinogenic. Dagoberto C. Brandão PHC Pharma Consulting
69
Polar Solvent Extract from the Euphorbiaceae Plants
E. tirucalli latex is agressive to the human body and therefore seen and recommended as something with wich any contact should be avoided; Thus, against all technical prejudice, the applicant verified that a specifc active fraction of na extract of said latex, as well as one or more compounds composing it, has effective anticancer action. Dagoberto C. Brandão PHC Pharma Consulting
70
Pre Clinical Pharmacology
Studies with mice inoculated with Ehrlich´s tumoral cells evaluated the antitumoral activity of the active fraction from the latex; antitumoral response statiscally significant with a dosage of 200mg/kg; Myelopoietic activity study: therapeutical effect originated from the direct citotoxic action on tumoral cells due to the activation of macrophages. Citotoxic effect: demonstrated in the significant reduction of the number of hematopoietic precursors (mainly with higher dosages).

Pre Clinical Toxicology
Preclinical studies revealed that Euphorbia tirucalli is toxic in mice and dogs can cause mucositis and hepatotoxicity in both; ACUTE TOXICITY BY 24 HS IN MICE(100, 300, 1000, 5000 mg/kg) Weight decrease without any change in demand for food and water, there were tremors, increased heart rate and reduction of body tonus; Produced low acute toxicity during the 14 days of observation (showed good tolerability and few toxic important effects). Dagoberto C. Brandão PHC Pharma Consulting
80
Pre Clinical Toxicology
TOXICITY IN REPETITIVE DOSAGES FOR 90 DAYS IN MICE (30, 100 and 300 mg/kg) There was a reduction in the consumption of rations and water in males, as well as in the number of leukocytes; There was low toxicity when evaluated under biochemical, hematological and histopatological parameters. Dagoberto C. Brandão PHC Pharma Consulting
81
Pre Clinical Toxicology
TOXICITY DURING 90 DAYS IN DOGS (0, 5, 10 and 20 mg/kg) Reduction of the consumption of food and corporal weight (dosage of 20 mg/ kg); Loss of fur, abdominal pain and diarrhea. Histopathology: incidence of 100% of the apoptotic figures, hyperplasia and hepatocites hypertrophy in exposure to 20mg/ kg and high incidence of mucous in the intestines of all exposed animals.
