Transferencia no radiativa de energía ES Energía Fluorescencia What is a tandem dye? A tandem dye is composed of two covalently conjugated fluorochromes with overlapping spectral characteristics. These characteristics allow the transfer of energy from one molecule to the other resulting in fluorescence emission at a longer wavelength. One molecule functions as the “donor” molecule, the other is the “acceptor” molecule. Current tandem dyes all use PE as the donor molecule. The schematic diagram demonstrates how energy is transferred from the donor molecule to the acceptor molecule. Energy applied to the dye in the form of light excites the donor molecule. The electrons move from the ground state to the excited state. Energy is released in the form of fluorescence when the electrons return to the ground state. When the excited states of the donor and acceptor molecule overlap, the released energy from the donor molecule is transferred to the acceptor molecule instead of returning to the ground state as fluorescence of the donor molecule. The electrons of the acceptor molecule then return to the ground state emitting fluorescence at the acceptor molecule wavelength. COMPENSATION IS INTENSITY DEPENDENT partially compensated
FL2 It is an absolute requirement that particles used for compensation be at least as bright as the antibody staining. If this condition is not met, antibodies whose fluorescence is brighter than the particles used for compensation will not be properly compensated. COMPENSATE INSTRUMENT USING STAINED CELLS
